- Take County Rd 46 N/William Floyd Pkwy, Longwood Rd, Yaphank Middle Island Rd and Canal Rd to 5225 Nesconset Highway.
From Northern State Parkway:
- Take eastbound Northern State Pkwy to NY-347/NY-454 E in Hauppauge.
- Follow NY-347 E to 5225 Nesconset Highway.
From Southern State Parkway:
- Take eastbound Southern State Pkwy and Sagtikos Pkwy to NY-347/NY-454 E in Hauppauge.

From Eastbound Sunrise Highway (NY 27-E):
- Follow NY-27 E to Sunrise Hwy South Service Rd in Bayport.
- Take exit 51 from NY-27 to E County Road 97/Nicolls Rd toward Blue Point/Stony Brook.
- Take Nicolls Rd and NY-347 E to 5225 Nesconset Highway.
From Montauk Westbound NY-27:
- Take NY-27 W/State Rte 27 W/POW/MIA Memorial Highway W to Upton.
- Continue on Longwood Rd. Take Canal Rd to NY-347 E in Terryville.
